Development and Application of a Validated Determination of Penta-O-galloyl-¥â-D-glucose in Extract of Evening-primrose Seeds as Dietary Supplement by High-Performance Liquid Chromatography

|
The purpose of this study is to develop and apply for determination of penta-O-galloyl-¥â-D-glucose (PGG), an essential marker compound in extract of evening-primrose seeds used as dietary supplement using by high-performance liquid chromatography (HPLC). Liquid chromatography was performed on a Capcellpak UG 120 column (4.6 mm ID ¡¿ 250 mm, 5 ¥ìm) using the gradient elution system developed in this study. According to the guidelines of the Association of Official Analytical Chemists for dietary supplements, the performance characteristics of HPLC were evaluated in terms of selectivity, linearity, precision, accuracy, recovery, limit of detection and limit of quantification. The calibration curve was highly linear (the coefficient of determination: 0.9999) within the concentration range of 0.42~26.88 ¥ìg/mL for PGG. The limit of detection and limit of quantitation of PGG in this method were 0.04 ¥ìg/mL and 0.13 ¥ìg/mL, respectively. This analytical method for determination of PGG as the marker compound was applied for domestic commercially available three extracts of evening-primrose seeds used as dietary supplements. The recoveries for these three fortified concentrations were 96.7~100.6%, 96.5~98.3% and 100.6~104.2%, respectively. It is expected that the validated HPLC analytical method developed in this study will be usefully applicable to determination of PGG in extract of evening-primrose seeds as dietary supplement for its quality control.
